An assay is a procedure where the concentration of a component part of a mixture is determined.

So, an enzyme assay determines the concentration of an enzyme. There are lots of ways to do this. One is to put in the enzyme's substrate and see how quickly it reacts. Assuming you know the relationship between enzyme concentration and reaction rate you can find the answer.
